Zavesca (miglustat) is a glucosylceramide synthase inhibitor used as monotherapy for the treatment of adult patients with mild/moderate type 1 Gaucher disease for whom enzyme replacement therapy is not an effective therapeutic option. It is not known if this oral therapeutic drug is safe and effective in children aged under 18 years.
Recommended Dosage: The recommended dosage for adult patients with type 1 Gaucher disease is one 100 mg capsule administered orally three times daily at regular intervals. If any dose is missed, the next capsule needs to be taken at the next scheduled time. It may be required to reduce the dose to a single 100 mg capsule once or twice daily in some patients because of side effects, such as tremors or diarrhea.
- Treatment with Zavesca 100 mg capsules can cause liver enzyme elevations, which may indicate liver damage. Healthcare providers (HCPs) should monitor liver function tests regularly during treatment.
- This therapeutic drug may be responsible for causing peripheral neuropathy, which can manifest as tingling, numbness, or pain in the hands and feet. Patients should report any symptoms of neuropathy to their healthcare provider (HCP) promptly.
- Treatment with Miglustat 100 mg oral capsules commonly causes gastrointestinal side effects such as diarrhea, nausea, and abdominal pain. Patients should maintain adequate hydration and report severe or persistent gastrointestinal symptoms to their healthcare provider.
- Miglustat may cause fetal harm if used during pregnancy. Women of childbearing age should use effective contraception while taking Zavesca and avoid pregnancy. Additionally, it is not known whether this therapeutic drug is excreted in human breast milk, so nursing mothers do not breastfeed during treatment.
- Miglustat may interact with certain medications, including CYP3A4 inducers and inhibitors, which could affect its effectiveness or increase the risk of adverse reactions. Patients should inform their healthcare provider (HCP) about all medications, supplements, and herbal products they are taking before starting treatment with Miglustat
