Ayvakit is a kinase inhibitor used for:
Gastrointestinal Stromal Tumor (GIST): to treat adults with unresectable or metastatic GIST harboring a platelet-derived growth factor receptor alpha (PDGFRA) exon 18 mutation, including PDGFRA D842V mutations.
Advanced Systemic Mastocytosis (AdvSM): to treat adult patients with AdvSM. Advanced Systemic Mastocytosis includes patients with mast cell leukemia, aggressive systemic mastocytosis, systemic mastocytosis with an associated hematological neoplasm (SM-AHN).
Recommended Dosage: The recommended dosage is 300 mg orally once daily in patients with GIST. Continue treatment until the disease is progressive or unacceptable toxicity occurs.
The recommended dosage is ayvakit 200 mg orally once daily in patients with AdvSM. Continue treatment with Ayvakit until the disease is progressive or unacceptable toxicity occurs.
Administer the tablets of Ayvakit orally on an empty stomach, at least 60 minutes before or 120 minutes after a meal. Avoid making up for a missed ayvakit dose within eight hours of the next scheduled dose. Do not repeat the dose in case vomiting occurs after taking Ayvakit dose but continue with the next scheduled dose.
- Severe intracranial hemorrhage may occur with avapritinib 300 mg treatment. Assess patients closely for the risk of intracranial hemorrhage including those with vascular aneurysm, thrombocytopenia or a history of intracranial hemorrhage or cerebrovascular accident within the earlier year.
- In patients with AdvSM, perform platelet count prior to initiating therapy; avapritinib 50 mg or any other dosage strength is not recommended in patients with Advanced Systemic Mastocytosis with platelet counts < 50 X 109.
- Cognitive adverse events can appear in patients receiving avapritinib 25 mg tablets. On behalf of the severity, withhold avapritinib and then resume at the same dose or at a decreased dose upon improvement, or permanently discontinue avapritinib.
- Avapritinib can cause fetal harm if administered to pregnant women. Males and females of reproductive potential should use effective contraception during treatment with avapritinib 200 mg and for 6 weeks following the final dose.
- Respective women should not breastfeed during treatment with tablets of Avapritinib and for 2 weeks following the last dose.
