Signifor LAR (pasireotide) is a somatostatin analog indicated for the treatment of:
- Patients with acromegaly who have had an inadequate response to surgery and/ or for whom surgery is not an option.
- Patients with Cushing’s disease for whom pituitary surgery is not an option or has not been curative.
Recommended Dosage: Signifor LAR must be administered by a trained healthcare professional only by intramuscular (IM) injection in the right or left gluteus immediately after reconstitution. This medication must never be administered intravenously.
- Acromegaly: The recommended initial dose for the treatment of acromegaly is 40 mg administered by intramuscular (IM) injection once every 4 weeks (every 28 days).
- Cushing’s Disease: The recommended initial dose for the treatment of Cushing’s disease is 10 mg administered by intramuscular (IM) injection once every 4 weeks (every 28 days).
- Signifor LAR can cause hyperglycemia (high blood sugar) and hypoglycemia (low blood sugar). Monitor blood glucose levels regularly during treatment, especially in patients with diabetes or at risk for glucose intolerance.
- Patients with hepatic impairment may experience elevated liver enzymes or liver damage while taking Pasireotide. Regular monitoring of liver function tests is recommended, and dosage adjustments may be necessary.
- Treatment with this medicinal product may suppress adrenal function, leading to adrenal insufficiency. Monitor patients for signs and symptoms of adrenal insufficiency, such as fatigue, weakness, and hypotension.
- Patients receiving this therapeutic drug may experience hypocortisolism, which can manifest as symptoms of adrenal insufficiency. Consider adjusting corticosteroid replacement therapy during treatment with this medicine.
- Patients with diabetes may experience worsening hyperglycemia while taking this medication. Monitor blood glucose levels closely and adjust diabetes management as needed.
- Pasireotide injection may cause fetal harm if administered during pregnancy. Use in pregnant women only if the potential benefit justifies the potential risk to the fetus. It is unknown whether the medicine is excreted in human breast milk; breastfeeding should be discontinued during treatment.
- Treatment with this medicine may interact with certain medications, including antidiabetic drugs and QT-prolonging agents. Use caution when co-administering Signifor LAR with other drugs and adjust dosages as necessary.
- Injection site reactions, such as pain, swelling, or bruising, may occur with Signifor LAR injection administration. Rotate injection sites and monitor for any local adverse reactions.
