Rozlytrek is a kinase inhibitor used for the treatment of adult patients with metastatic non-small cell lung cancer (NSCLC) whose tumors are ROS1-positive.
Rozlytrek is also used for adult and pediatric patients aged 12 years and older with NTRK fusion-positive solid tumors.
Recommended Dosage: The recommended dosage for ROS1-Positive Non-Small Cell Lung Cancer is 600 mg orally once daily with or without food until disease progression or unacceptable toxicity.
The recommended dosage for NTRK Gene Fusion-Positive Solid Tumors in adults is 600 mg orally once daily, and for Pediatric Patients aged 12 years and older (Adolescents) with Body Surface Area (BSA) greater than 1.50 m2 dose is 600 mg; for BSA 1.11 to 1.50 m2 dose is 500 mg and for BSA 0.91 to 1.10 m2 dose is 400 mg. Take this medicine orally once daily with or without food until the disease is progressive or unacceptable toxicity occurs.
- Congestive heart failure (CHF) may occur. Assess left ventricular ejection fraction (LVEF) before initiating treatment with entrectinib 100 mg capsule or any other dosage strength in patients with symptoms or known risk factors for CHF.
- Monitor patients for clinical signs/symptoms of CHF, including edema and shortness of breath. For patients with myocarditis, with or without a reduced ejection fraction, MRI or cardiac biopsy may be needed to make the diagnosis.
- A broad spectrum of CNS adverse reactions may occur in patients with rozlytrek 100 mg capsule, including mood disorders, cognitive impairment, dizziness, and sleep disturbances. Patients should not drive or operate hazardous machinery in case they are experiencing CNS adverse reactions.
- Entrectinib increases the probability of fractures. Promptly assess patients with signs/symptoms (e.g., changes in mobility, pain, deformity) of fractures. There are no findings on the effects of the entrectinib capsule on the healing of known fractures and the probability of future fractures.
- Increased AST of any grade may occur. Monitor liver tests, including AST and ALT, every 14 days during the initial month of treatment, then monthly after that. Withhold or permanently discontinue Entrectinib therapy on behalf of the severity. If withheld, resume therapy with Entrectinib at the same or reduced dose.
- Patients may experience hyperuricemia. Assess serum uric acid levels before initiating treatment with Rozlytrek capsule and periodically during treatment. Also, monitor patients for signs and symptoms of hyperuricemia. Start treatment with urate-lowering drugs and withhold Rozlytrek for signs/symptoms of hyperuricemia.
- Findings and its mechanism of action suggest that Entrectinib can cause fetal harm if administered to a pregnant woman. Also, mothers should avoid breastfeeding during treatment with Entrectinib and for 7 days after the final dose.
