- Prolia is a RANK ligand (RANKL) inhibitor used for the treatment:
- of postmenopausal females with osteoporosis at high risk for fracture
- to increase bone mass in males with osteoporosis at high risk for fracture
- of glucocorticoid-induced osteoporosis in males and females at high risk for fracture
- to increase bone mass in males at high risk for fracture receiving androgen deprivation therapy for nonmetastatic prostate cancer
- to increase bone mass in females at high risk for fracture receiving adjuvant aromatase inhibitor therapy for breast cancer
Recommended Dosage: The recommended dose of Prolia injection is 60 mg administered as a single subcutaneous injection once every 6 months. To administer Prolia, inject it subcutaneously into the upper arm, upper thigh, or abdomen. Patients should also take 1000 mg of calcium and at least 400 IU of vitamin D daily.
If a dose of Prolia is missed, give the injection as soon as possible. Then, continue injections every 6 months from the date of the last one.
- Severe hypocalcemia can occur due to Prolia usage. Prior to initiating Prolia therapy, it’s essential to address any existing hypocalcemia. Additionally, assess patients for chronic kidney disease mineral-bone disorder and regularly monitor serum calcium levels.
- Since Prolia and Xgeva both contain denosumab, patients who are being treated with Prolia 60 mg injection should avoid receiving Xgeva.
- Prolia may cause severe hypersensitivity reactions, including anaphylaxis. Symptoms can include low blood pressure, difficulty breathing, tightness in the throat, itching, and hives. If an allergic reaction occurs, seek medical attention promptly and stop using Prolia.
- After stopping Prolia treatment, there is an increased risk of fractures, particularly multiple vertebral fractures. These new fractures can occur as soon as 7 months (on average 19 months) after the last dose. Prior vertebral fracture increases the likelihood of experiencing multiple vertebral fractures after stopping Prolia. It’s important to carefully consider the benefits and risks for each individual before starting Prolia treatment.
- Patients taking Prolia may experience severe and sometimes debilitating bone, joint, and/or muscle pain. Symptoms can appear as early as one day or as late as several months after starting treatment. If severe symptoms occur, it may be advisable to discontinue Prolia use.
