Recommended Dosage: After an initial titration, the recommended dosage of aduhelm injection is 10 mg/kg. It is administered as an intravenous (IV) infusion over approximately 60 minutes every four weeks and at least 21 days apart.
In case of missed infusion, resume administration at the same dose as quickly as possible. Infusions are to be administered every four weeks and at least 21 days apart.
- Aduhelm 170 mg therapy may cause amyloid related imaging abnormalities-edema (ARIA-E), which can be observed on MRI as brain edema or sulcal effusions, and amyloid related imaging abnormalities-hemosiderin deposition (ARIA-H), which includes microhemorrhage and superficial siderosis.
- Obtain recent (within 12 months) brain MRI prior to starting treatment. The safety of this medicine in patients with any pre-treatment localized superficial siderosis, 10 or more brain microhemorrhages, or with a brain hemorrhage of more than 1 cm within 12 months of therapy initiation has not been established.
- Brain MRIs need to be obtained before the 7th infusion (1’st dose of 10 mg/kg) and 12th infusion (6’th dose of 10 mg/kg) of aduhelm 300 mg to evaluate for the existence of asymptomatic ARIA. For patients with radiographic findings of ARIA, enhanced clinical vigilance is needed.
- Urticaria and angioedema were reported in 1 patient in the placebo-controlled time-span of Studies 1 and 2, and emerged during the infusion. Quickly discontinue the infusion upon the initial observation of any symptoms consistent with a hypersensitivity reaction, and start suitable therapy.
