Drug Overview:
Sotorasib is supplied under the brand name Lumakras. The Food and Drug Administration (FDA) has approved this prescription drug to treat non-small cell lung cancer (NSCLC) that’s locally advanced or metastatic. It’s used in adult patients who:
- have NSCLC that has an abnormality in a gene named KRAS G12C, and
- have previously tried a systemic treatment
Sotorasib comes as an oral tablet in two dosage strengths 320 mg and 120 mg. It falls under a drug class known as RAS GTPase inhibitors. At this time, the medicine Sotorasib only exists as a brand-name drug, not a generic.
Dosage and Administration:
The recommended dosage of Sotorasib is 960 mg (three 320 mg tablets or eight 120 mg tablets) orally once daily until there is evidence of disease progression or the emergence of unacceptable toxicity. Take the daily dose of Sotorasib at the same time each day with or without food. Swallow tablets whole. Do not chew, crush, or split tablets. If a dose of Sotorasib is missed by more than 6 hours, take the next dose as prescribed the next day. It is not advisable to take 2 doses at the same time to make up for the missed dose. If vomiting occurs after taking Sotorasib, do not take an additional dose. Take the very next dose as prescribed the next day.
Potential Side Effects: The commonly reported side effects of Sotorasib with incidence of ≥ 20% are:
- diarrhea
- musculoskeletal pain
- nausea
- fatigue
- hepatotoxicity,
- and cough
